Race car Lightning McQueen (Owen Wilson) returns home to Radiator Springs after winning a fourth Piston Cup championship, and reunites with best friend Mater (Larry the Cable Guy) and girlfriend Sally Carrera (Bonnie Hunt). Former oil tycoon Miles Axlerod (Eddie Izzard), now a green power advocate, announces a three-race series called the "World Grand Prix", as a means to promote Allinol biofuel. Although McQueen initially refuses to participate, Italian formula race car Francesco Bernoulli's (John Turturro) challenge, as well as Mater's intervention, leads McQueen to enter the race. Despite the fact that McQueen never took Mater with him to any of his Piston Cup races, at Sally's urging, he agrees to take Mater with him to the World Grand Prix.

Meanwhile, an group of "lemon" cars led by Professor Zündapp (Thomas Kretschmann) and an unknown mastermind owns the largest untapped oil reserves in the world. They secretly plot to secure their oil profits by using a weapon disguised as a television camera that would ignite the Allinol fuel in targeted race cars during the World Grand Prix; with no apparent cause for the explosions, the public would doubt the fuel's safety and depend further on the group's oil. British spy cars Finn McMissile (Michael Caine) and Holley Shiftwell (Emily Mortimer) attempt to foil their plot. They attempt to rendezvous with American spy car Rod "Torque" Redline (Bruce Campbell) at a World Grand Prix promotional event in Tokyo in order to receive information about the mastermind; however, Redline is cornered by Zündapp's henchmen and passes his information to an unsuspecting Mater before being captured and subsequently killed. As a result, Shiftwell and McMissile mistake Mater as their American contact; Mater in turn thinks Shiftwell's attempted rendezvous is flirting.

When the Tokyo race begins, McMissile and Holley move to protect Mater from Zündapp's henchmen. While obliviously evading the henchmen, Mater inadvertently gives McQueen bad advice which causes him to lose the race. Meanwhile, Zündapp uses the weapon to ignite the Allinol in several race cars. Angry at Mater's embarrassing behavior as well as causing his loss, McQueen berates Mater, who decides to return to Radiator Springs. McMissile, who still believes Mater is an American spycar, picks Mater up at the airport and drafts him into foiling Zündapp's plot.
In Italy, the site of the second race of the Grand Prix, a disguised Mater infiltrates the criminals' meeting and discovers Zündapp's plan. Zündapp's henchmen, meanwhile, use their weapon on most of the cars in the race, and McQueen and Bernoulli end up in first and second place, respectively. With the Allinol fuel under suspicion, Miles Axlerod decides to suspend its use for the final race in England; however, McQueen decides to continue using the fuel. The criminals then decide to have McQueen killed in the next race. Mater reveals himself by accident and rushes to warn his friend, but is captured along with McMissile and Shiftwell before he is able to do so.

Mater wakes up tied up inside Big Bentley along with McMissile and Shiftwell, minutes away from being crushed by its gears. The last race begins and the criminals use the weapon on McQueen, but surprisingly, nothing happens. Mater learns that a bomb was put into Lightning's pit crew area, set to explode when McQueen stopped there, and he manages to escape to warn his friends. McMissile and Shiftwell succeed in freeing themselves as well, but find that the bomb was planted on Mater. They warn Mater about the bomb just in time, and Mater flees to protect his friends, with McQueen, unaware of the situation, in pursuit.

Professor Zündapp tries to detonate the bomb remotely only to find that the device is out of signal range. He then sends his henchmen to kill McQueen and Mater, but they are foiled by the combined efforts of McMissile, Shiftwell, and the Radiator Springs residents. McMissile orders Zündapp to deactivate the bomb in Mater's body, but Zündapp claims that the bomb is voice activated and only the one who installed it is able to do so. Mater then figures out that the mastermind behind the Allinol plot is Miles Axlerod himself. Axlerod confirms Mater's suspicion when Mater confronts him and Axelrod is able to deactivate the bomb.

For stopping Axlerod's plans, Mater is knighted by the Queen and returns home with his friends, where the cars from the Grand Prix take part into their own unofficial Radiator Springs Grand Prix. Fillmore reveals that Sarge replaced Lightning's Allinol before the last race with Fillmore's own organic fuel mixture, which prevented Lightning from being affected by the weapon. McMissile and Shiftwell visit Mater and invite him to join them in another secret mission, but he declines, claiming that he is in the place he should be. wikipedia
